It is a traditional cell tissue rapid lysate with a moderate degree of lysis, including the cell membrane, cell cytoplasm and cell nucleus components of animal cells.
Introduction
RIPA Lysis Buffer is a traditional rapid lysis buffer of cellular tissues that is primarily used for soluble proteins extracted from animal tissues and animal cells. There are many kinds of formulas, According to the strength of the lysis buffer, it can be roughly divided into three categories: enhanced, medium and weak.
This product is a lysis buffer that uses classical tissue cells to rapidly lyse and obtain total protein, with a strong degree of lysis, and its lysis strength is greater than that of RIPA lysis buffer (medium) and NP-40 lysis buffer. The protein samples obtained by lysis can be used for conventional PAGE, Immunol Precipitation (IP), Western, etc. This product contains protease inhibitor ingredients that effectively inhibit protein degradation.
Protein samples lyzed with RIPA lysis buffer can be using the BCA Protein Concentration Determination Kit. However, due to the higher concentration of detergent, the protein concentration of the sample obtained by the lysis of this lysate cannot be determined by Bradford method.

Q: Transparent gelatinous substances often appear in the lysate of RIPA lysis buffer?
A:
This jelly is a complex containing genomic DNA, etc., which is a normal phenomenon, and can be selected as follows according to different experiments:
1) When the target protein is not tightly bound to the genomic DNA, the lysate can be centrifuged directly, and the supernatant is taken for subsequent experiments;
2) When the target protein is particularly tightly bound to the genomic DNA, the transparent gel can be broken up by ultrasonic treatment, and the supernatant can be taken by centrifugation for subsequent experiments;
3) If some common transcription factors (such as NF-kappaB, p53, etc.) are detected, these transcription factors can usually be detected without sonication.
