Introduction
BCA protein quantification is currently the most widely used protein quantitation method. This product is an enhanced BCA protein quantification kit developed on the basis of BCA protein quantitative, which realizes the simplicity, high stability, high sensitivity and high compatibility of protein concentration determination. Compared to ordinary BCA protein quantification kits, the color development speed is faster, and the same sample can be incubated for a short time for absorbance determination.
This product has a good linear relationship in the concentration range of 20~1000 μg/mL. The lower limit of detection concentration can reach 10 μg/mL, and the minimum detected protein amount can reach 0.2 μg. The volume of the sample to be tested is 1~20 μL.
This product can be compatible with 5% SDS, 5% Triton X-100, 5% Tween 20(60, 80), affected by chelating agents and slightly higher concentrations of reducing agents, but it is necessary to ensure that there is no EGTA, EDTA is less than 10 mM, deoxidizer DTT is less than 1 mM, β-Mercaptoethanol less than 1 mM. If the sample diluent or lysate itself has a high background and the BCA method is not applicable, please use other methods to determine the protein concentration.

Q1:测定标准曲线时,发现随着标准品浓度的增加,其吸光值或颜色没有明显变化?

可能是所使用的标准品稀释液中含有严重干扰BCA法测定蛋白浓度的物质。建议将稀释液做本底对照,查看吸光值。
Q2:每次测定蛋白浓度时都需要做标准曲线吗?
A2:
建议每次测定时都做标准曲线。因为BCA法测定蛋白浓度时,颜色会随着时间的延长不断加深,并且显色反应会因温度升高而加快。所以除非精确控制显色反应的时间和温度,否则每次测定蛋白浓度都应该做标准曲线。
