Introduction
T4 DNA Ligase is expressed from E. coli carrying the T4 DNA ligase gene. The enzyme can catalyze the formation of phosphodiester bonds between 5'-phosphate groups and 3'-hydroxyl groups between double helix DNA or RNA. The enzyme can repair single-stranded nicks between double-stranded DNA, RNA or DNA/RNA complexes, and can ligate DNA fragments with cohesive or blunt ends, but is inactive for single-stranded nucleic acids. T4 DNA Ligase requires ATP as a cofactor and completes the cohesive end ligation reaction in as little as 10 minutes at room temperature.
