The 500 bp and 1,500 bp fragments are brighter than the other fragments in 250 bp DNA marker and concentration of these indicator fragments are 150 ng/5 μL, other fragments contains ~50 ng/5 μL.

Q1：DNA Marker/Ladder无法正常分离？

A1：

1) 琼脂糖凝胶浓度不合适。选择合适的凝胶浓度；

2) 核酸染料质量不佳影响大分子量DNA染色效果。选择质量及兼容性较好的核酸染料（目录号：GL802）；

3) 缓冲液陈旧导致缓冲能力下降。及时更换新鲜缓冲液；

4) 缓冲液配制有误。检查配制方案。

 

Q2：DNA Marker/Ladder条带弯曲？

A2：

1) 电压过高。电泳时电压建议5~10 V/cm；

2) 电泳过程中电压不稳。及时检修电泳仪；

3) 胶孔不整齐或上样时枪头插入胶中。待凝胶完全凝固后垂直拔下梳子、上样时避免枪头插入凝胶中；

4) 缓冲液陈旧导致缓冲能力下降。及时更换新鲜缓冲液。

 

Q3：DNA Marker/Ladder模糊、弥散？

A3：

1) DNA Marker/Ladder降解。上样时注意更换枪头；

2) 核酸染料质量不佳影响大分子量DNA染色效果。选择质量及兼容性较好的核酸染料(目录号：GL802)；

3) 缓冲液陈旧导致缓冲能力下降。及时更换新鲜缓冲液；

4) 缓冲液配制有误。检查配制方案。
